Production and optimization of laccase and lignin peroxidase by newly isolated Pleurotus ostreatus LIG 19

Abstract

In the present study, a lignolytic fungus was isolated from Chennai which was able to produce laccase and lignin Peroxidase. The organism was identified as Pleurotus ostreatus using 28 S rRNA sequence analysis. The parameters like pH, temperature, carbon source, nitrogen source were optimized for better production of lignin peroxidase and laccase. The mass production of the enzymes was carried out using optimized conditins and partial purification of lignin peroxidase and laccase was employed by ammonium sulphate precipitation and dialysis. The partially purified lignin peroxidase and laccase was characterized and the protein profile was analyzed in SDS-PAGE.  Keywords: Lignolytic enzymes, Guaiacol, 28S rRNA sequence analysis, Pleurotus ostreus
